Figure 3From: Novel action of apolipoprotein E (ApoE): ApoE isoform specifically inhibits lipid-particle-mediated cholesterol release from neuronsEM mediated cholesterol and PC release from cultured neurons. Neurons labeled with 37 Bq/ml [14C]acetate were cultured for 3 days at 37°C. These neurons were washed in DMEM three times and further incubated in DMEM for 1 h (a) or various times (b) in the presence of EM at various PC concentrations. The conditioned media were then collected and the amounts of [14C]-labeled cholesterol and PC were determined. (a) EM-induced cholesterol (○) and PC (●) release from cultured neurons occurs in an EM-PC-dose-dependent manner. (b) The time-dependent release of cholesterol (○) and PC (●) from cultured neurons in the presence of EM at PC concentration of 5 μg/ml is shown. Data are means ± S.E. of four samples. *p < 0.0001 and **p < 0.001 vs PC at 0 ng/ml and time at 0 h. Four independent experiments showed similar results.Back to article page
