Knockdown of OSBP1 expression using siRNA increased secreted levels of Aβ40 and Aβ42 and the steady-state levels of CTFβ. Cells were transfected with the 3 siRNA sequences as described in Methods. Non-targeting siRNA (NT) and siRNA against APP and BACE1 were used as controls. A, Media samples were assayed for Aβ40 and Aβ42 by ELISA. B, Lysates from HEK-APPNFEV cells were separated by SDS-PAGE and immunoblotted with an antibody to the APP C-terminus for detection of full length APP and APP-CTFs, and to actin as loading control.
