B, Immunoblot analysis of UPR markers in brainstem of Hup23 mice. Seventy five ÎĽg of total protein lysates of brainstem of Hup23 and non-transgenic littermates were fractionated on 4-20% SDS-PAGE gels and immunoblotted using antibodies against the indicated proteins. Hsc70 and GAPDH levels served as loading controls. Lysates from mouse embryonic fibroblasts (MEF) exposed overnight to 2 ÎĽg/ml tunicamycin (Tm) were analyzed on the same gels as positive control for the activation of the UPR.
