Western blot analyses of αSyn_RT-QuIC products from OM-seeded reactions. Representative Western blot images showing the presence of PK resistant bands in αSyn_RT-QuIC end products generated by PD (n = 8) (a) and MSA-P (n = 8) samples (b). Conversely, no bands were detected in αSyn_RT-QuIC end products seeded with HS (n = 8) (c) and MSA-C (n = 8) (d). Statistical analysis revealed that PD generated products were significantly less resistant to PK digestion compared to those generated by MSA-P (Mann-Whitney test p = 0.0104). Densitometric analyses have been performed on Western blot images of αSyn_RT-QuIC products generated from PD (n = 13), MSA-P (n = 20), MSA-C (n = 10) and HS (n = 11) OM samples at ITA-lab (see Supplementary Fig. 3a).
