Enhancement of glutamate-induced excitotoxicity by tau does not depend on GluN2B/2B–containing NMDARs. a, b Glutamate-induced neurotoxicity in Mapt −/− neurons transduced with lentiviral vectors encoding GFP-P2A-mTauWT or GFP-P2A was assessed as in Figs. 1 and 2. For 1 h before and throughout the glutamate exposure, neurons were treated with a vehicle or b Ro 25–6981 (1 μM). Numbers of independent experiments (n) with cumulative well numbers per condition in parentheses: a 3 (23–24) and b 4 (28–32). When comparing mean differences across all doses within any given panel, a one-sided, one-sample t-test revealed significant differences between experimental and control conditions in (a: P < 0.05) and (b: P < 0.05)
